Özet

Chitinase is an enzyme that breaks down chitin, a complex carbohydrate found in the cell walls

of fungi, exoskeletons of arthropods (such as insects and crustaceans), and some other organisms.

Chitinase was extracellularly produced from Jiangella alba in ISP2 liquid medium (4 g/L yeast extract,

10 g/L malt extract, and 4 g/L glucose, pH 7.0 ± 0.2) supplemented with colloidal chitin (10 g/L) for

7 days at 32 °C (Ding vd., 2023; Adıgüzel vd., 2023). The enzyme was retrieved from the culture by

centrifugation (10.000 x g, 10 min) and then subjected to ammonium sulfate precipitation with 80%

saturation. Precipitated enzyme dissolved in 50 mM phosphate buffer (pH 7.0) was used as crude

chitinase. Enzyme activity was assayed by the dinitrosalicylic acid (DNS) method using colloidal chitin

as substrate (Yahiaoui 2019). Crude chitinase was active at a temperature range from 20 to 50 °C and

exhibited maximal activity at 45 °C. Its maximum activity was recorded in 50 mM phosphate buffer at

pH 6.0. The enzyme lost its activity about 28, 43, and 50% after pre-incubated at 40, 45, and 50 °C for an

hour. Maximum chitinase activity was observed in the presence of NaCl. Elevated concentration of NaCl

caused a gradual decrease in the chitinase activity. Adding Mg2+ and Ca2+ at a five mM final concentration

to the reaction mixture increased the activity of crude chitinase. On the other hand, a significant decrease

in the enzyme activity was observed when the Cu2+ was introduced to the reaction mixture. In addition,

5 mM Triton X-100 slightly increased the activity of chitinase. Results showed that the chitinase from

Jiangella alba has a potential for use in various industrial applications such as biological treatment of

seafood, textile processing, drug development, and material design for packaging. In addition, it may

be used as a biocontrol agent.
